A fully integrated isotachophoresis with a programmable microfluidic platform

Conventional isotachophoresis (ITP) can be used for pre-concentration of a single analyte, but preconcentration multiple analytes is time consuming due to handling and washing steps required the extensive buffer optimization procedure. In this work, we present programmable microfluidic platform (PMP) demonstrate fully automated ITP analytes. By interfacing PMP with ITP, selection repetitive procedures were automated. Using lifting-gate microvalve technology, consisting two-dimensional array was designed fabricated seamless integration an chip. The basic liquid manipulation such as metering, mixing, selecting, delivering, prime run ITP. Initially, performances channel validated individually by estimating volume per pumping cycle preconcentrating Alexa Fluor 594 appropriate trailing (TE) leading (LE) buffers, respectively. After confirming functions, autonomous demonstrated using (Pacific blue, 594, 488). optimal combination determined performing runs three different TEs (borate, HEPES, phosphate buffers) concentrations Tris-HCl LE. We found that 40 mM borate 100 successfully preconcentrated all during run. integrated PMP-ITP system simplify overall validation various biological chemical target samples. Furthermore, incorporating analytical tools interconnect PMP, it provide high sample aid in downstream analysis.
